Glycan Analysis Workflow

Most of the functional properties of glycoprotein are dependent on the glycosylation pattern and occurrence of glycosylation at the probable sites. The N-glycosylation profile of the glycoprotein can be generated by cleaving the Asp-linked N-glycan from protein by PNGase F and analyzed by HILIC–MS technique. The linkage of each glycan form can be confirmed through the digestion with specific exoglycosidases and the confirmed through HILIC-MS and MS/MS. The column and system are equilibrated with dextran ladder before each set of HPLC runs which also provide the GU value of each glycoforms and can act as a standalone analysis technique. Global Proteomic Profiling Workflow

Proteins are the key functional actors in all cellular processes, and diseases almost always manifest at the expressed level of proteins. The analysis of all expressed proteins (the proteome), for example healthy and diseased tissues, is definitely the most applicable approach to uncover key players, which are often named as biomarkers or functionally related protein signatures. For the service of protein analysis, we will apply mass spectrometry (MS)-based “shotgun” proteomics to measure the protein sequence and concentration. Sequencing grade protein enzyme is applied for the digestion of entire set of proteins. Subsequently, all digested peptides are analyzed by using UHPLC MS/MS. Protein sequence will be analyzed by MASCOT and SEQUEST software. We can help you to identify the protein ID of your interest, and discover more proteins from all kinds of samples.
